Getting to grips with migraine: Researchers identify some of the biological roots of migraine from large-scale genome study

June 23, 2013 ? Migraine is an extremely difficult disorder to study. Between episodes, the patient is basically healthy, making the underlying pathology very difficult to uncover. In the largest migraine study, an international team of researchers have identified genetic regions linked to the onset and susceptibility of migraine.

In the largest study of migraines, researchers have found 5 genetic regions that for the first time have been linked to the onset of migraine. This study opens new doors to understanding the cause and biological triggers that underlie migraine attacks.

The team identified 12 genetic regions associated with migraine susceptibility. Eight of these regions were found in or near genes known to play a role in controlling brain circuitries and two of the regions were associated with genes that are responsible for maintaining healthy brain tissue. The regulation of these pathways may be important to the genetic susceptibility of migraines.

Migraine is a debilitating disorder that affects approximately 14% of adults. Migraine has recently been recognized as the seventh disabler in the Global Burden of Disease Survey 2010 and has been estimated to be the most costly neurological disorder. It is an extremely difficult disorder to study because no biomarkers between or during attacks have been identified so far.

"This study has greatly advanced our biological insight about the cause of migraine," says Dr Aarno Palotie, from the Wellcome Trust Sanger Institute. "Migraine and epilepsy are particularly difficult neural conditions to study; between episodes the patient is basically healthy so it's extremely difficult to uncover biochemical clues.

"We have proven that this is the most effective approach to study this type of neurological disorder and understand the biology that lies at the heart of it."

The team uncovered the underlying susceptibilities by comparing the results from 29 different genomic studies, including over 100,000 samples from both migraine patients and control samples.

They found that some of the regions of susceptibility lay close to a network of genes that are sensitive to oxidative stress, a biochemical process that results in the dysfunction of cells.

The team expects many of the genes at genetic regions associated with migraine are interconnected and could potentially be disrupting the internal regulation of tissue and cells in the brain, resulting in some of the symptoms of migraine.

"We would not have made discoveries by studying smaller groups of individuals," says Dr Gisela Terwindt, co-author from Leiden University Medical Centre. "This large scale method of studying over 100,000 samples of healthy and affected people means we can tease out the genes that are important suspects and follow them up in the lab."

The team identified an additional 134 genetic regions that are possibly associated to migraine susceptibility with weaker statistical evidence. Whether these regions underlie migraine susceptibility or not still needs to be elucidated. Other similar studies show that these statistically weaker culprits can play an equal part in the underlying biology of a disease or disorder.

"The molecular mechanisms of migraine are poorly understood. The sequence variants uncovered through this meta-analysis could become a foothold for further studies to better understanding the pathophysiology of migraine" says Dr K?ri Stef?nsson, President of deCODE genetics.

"This approach is the most efficient way of revealing the underlying biology of these neural disorders," says Dr Mark Daly, from the Massachusetts General Hospital and the Broad Institute of MIT and Harvard. "Effective studies that give us biological or biochemical results and insights are essential if we are to fully get to grips with this debilitating condition.

"Pursuing these studies in even larger samples and with denser maps of biological markers will increase our power to determine the roots and triggers of this disabling disorder."

U.S. boss held by Chinese workers awaits outcome

BEIJING (AP) ? An American executive held in his Beijing medical supply plant by angry workers said Tuesday's he's waiting for his lawyers to arrive to resolve a compensation dispute that highlights tensions in China's labor market.

Chip Starnes was enduring a fifth day of captivity at the factory in the capital's northeastern suburbs that makes products for Coral Springs, Florida-based Specialty Medical Supplies.

About 100 workers are demanding generous severance packages identical to those offered 30 workers being laid-off from the company's plastics division. The demands followed rumors the entire plant was being closed, despite Starnes' assertion the company doesn't plan to fire the others.

"There's been no solution is terms of anything between us and them," Starnes told The Associated Press from his office. He said he was waiting for his lawyer to arrive and then would "start to work on some sort of solution to the issue one way or another."

A local union official representing the workers in talks with Starnes, Chu Lixiang, said the workers were demanding the portion of their salaries yet to be paid and a "reasonable" level of compensation before leaving their jobs. Neither gave details on the amounts demanded.

Chu said Starnes hadn't paid the workers for two months. She said they feared the plant was closing and that he would run away without paying severance.

Starnes said that since Saturday morning, about 80 workers had been blocking every exit around the clock and depriving him of sleep by shining bright lights and banging on windows of his office.

The standoff points to long-ingrained habits among Chinese workers who are sometimes left unprotected when factories close without severance or wages owed.

Such incidents have been rarer as labor protections improve, although disputes still occur and local governments have at times barred foreign executives from leaving until they are resolved.

Starnes, 42, said he'd been coerced into agreeing to meet workers' demands by Tuesday.

Starnes said the company had gradually been winding down its plastics division, planning to move it to Mumbai, India. He arrived in Beijing a week ago to lay off the last 30 people. Some had been working there for up to nine years, so their compensation packages were "pretty nice," he said.

Some of the workers in the other divisions got wind of this, and, coupled with rumors that the whole plant was moving to India, started demanding similar severance packages on Friday, Starnes said.

Funeral plans set for actor James Gandolfini

NEW YORK (AP) ? The funeral for James Gandolfini will be held Thursday at the Cathedral Church of Saint John the Divine in New York City.

An HBO spokeswoman speaking on behalf of Gandolfini's family says the funeral is scheduled for 10 a.m.

The 51-year-old star of "The Sopranos" died Wednesday in Rome. Family spokesman Michael Kobold says Gandolfini died of a heart attack.

Port Authority of New York & New Jersey spokesman Steve Coleman tells the Star-Ledger of Newark that a Signature Airlines flight carrying Gandolfini's body arrived Sunday night at Newark Liberty International Airport from Rome. The body was taken from the airport around 11:20 p.m. but Coleman did not say where it went.

The actor had been headed to Sicily to appear at the Taormina Film Festival, which paid tribute to him Saturday.

How To Find The Right Teacher In Martial Arts | Content for Reprint

It goes without saying that not all teachers (of any subject) are equal and that is probably multiplied to the nth degree in the martial arts which doesn't allow for standardization to quite the same degree as other subjects might. There are of course ongoing attempts at standardization and classification but the martial arts are still very much of a melting pot and one is often left wondering who is teaching what and where did it come from.

Enter the beginner?

Given this prodigious output of styles and teachers, how is a beginner, who knows virtually nothing about the martial arts?how is such a person to find the right teacher?

And is that the right question?

Maybe there is no "right" teacher. Maybe the right teacher for me is not the right teacher for you.

So it can get a bit complicated?

But I'm getting ahead of myself here. Questions such as these simply do not arise for most people. They join their neighborhood club, pay their fees, get graded a few times, get a nice colored belt maybe even a trophy or two, tell their friends they are a brown belt in such and such a style and?that is all they want. And I'm not here to argue with that, but?

If you're real serious, if martial arts is your reason for living, you will need to find the Right Teacher and that means that you will have to do some serious searching. Both internally and externally.

Why?

Because the Right Teacher will not appear miraculously on the horizon when you first decide to be the next Bruce Lee. And so, almost by default, beginners tends to think of their first teacher as THE RIGHT TEACHER! Which is understandable because they don't know any better. They have nothing to compare their first teacher to, so the first teacher is THE BEST. The beginner is totally convinced of that.

However, as time moves on the beginner may begin to experience some disquiet. Some things don't add up. The first teacher may be a bit overweight, or not as fit as he could be. Maybe he doesn't answer questions satisfactorily?but hey! nobody's perfect and all things considered the first teacher is still The Best, and that's all there is to it, right? Nonetheless there is talk...there is talk of another teacher down the road who is (blasphemy!) even better. The beginner puts all those preposterous notions aside of course, but...

There are now some cracks in the facade.

Not fatal, but it is through these cracks that the doubts seep in.

That, at least, is what happened to me. I was absolutely convinced that my first teacher was the best, but?I checked out the new teacher down the road (no harm in that, is there?). Then I joined up and started the cycle all over again. And then again and again and again?

I tried judo, ju-jutsu, Hsing-i, Pa-kua, Tai chi, Shaolin and a number of other styles the names of which I now forget. Nothing changed. The teachers were all promising to begin with and, truth be told, they were all quite capable in their own way and I learned a lot from each and every one of them but they were only technicians. They knew the techniques, and that is all they knew. And I wanted more, so I moved on. Just what I wanted I was still not sure of?

An astute reader will now see the obvious. One's Search, one's Quest, is part of one's over-all Training. A very important part. Why?

Because it is all part of a learning curve. You are learning not only new techniques but you are learning something infinitely more important: the Art of Discernment! The Art of separating the wheat from the chaff. The Art of separating the Real from the False?

And yes, it is also the Art of recognizing the Real Teacher when he or she finally appears.

You see, that is the secret. Finding the right teacher is only the half of it. Recognizing them to be such is by far the more difficult and important half.

Let me ask you a question: how often does a student find the right teacher and continue to walk on by and not know the difference?

It happens more often than you might think! I've seen it again and again and again!

The thing to bear in mind is that the Right Teacher may not be (and usually isn't) the Hollywood stereotype. The Right Teacher may be an unassuming little fellow who runs a little corner store, is disarmingly self-effacing and has maybe one or two students who are equally unimpressive. In short this is not really the sort of person who matches your mental image of a deadly warrior. Or he may be crude and rude and obnoxious. And (blasphemy again!) he may not have your best interests at heart!

In other words, not all Masters fit the Hollywood mold! You really do have to keep your wits about you on this because the Right Teacher will confound your expectations nearly every time!

The "secret" then is to keep training, keep looking and to develop the Power of Discernment so that you can see below superficial appearances. (This will stand you in good stead not only in your search for a teacher and but also when you are in a fight!)

There is a saying that when the student is ready, the Teacher will appear. And that will happen when the student has developed a keen sense of discernment.*

*Author's note: by discernment I do not mean cynicism. The "been there, done that" attitude will not help you find a Teacher and if perchance you do, that Teacher will not be interested in teaching you. Real discernment has a quality of humility: you don't know the answer, you don't even know if there is an answer, but you will not stop searching.

Says cross-discipline learning is key

SAN RAFAEL ? Dominican University of California today said Sam Beldona will become the new dean of its School of Business and Leadership on July 31.

Mr. Beldona has most recently served as associate dean of the Graduate School of Business at Bryant University in Rhode Island, a role he has held since 2011. He also chaired the school?s department of management from 2007?2011.

Partnerships across academic disciplines was also a priority for business programs under his leadership, Mr. Beldona said in the announcement.

?What attracted me to Dominican was the compelling vision of the senior management team, as well as the energy and enthusiasm that the faculty and administrators demonstrated for new ideas, cross-disciplinary programs and global connections,? he said.

As an instructor, Mr. Beldona?s work has focused on undergraduate and graduate-level topics in international business and global finance. He has helped to create programs that expanded Bryant University?s global reach, including international study and intercultural programs.

Mr. Beldona helped to launch undergraduate majors in human resource management, entrepreneurship and global supply-chain management at Bryant. At the graduate level, he helped to build connections between regional companies and faculty to develop high-level specialty programs in global supply-chain management, international business and global finance.

Students were required to participate in several hands-on courses each year, including business pitches, service learning, internships and competitions. Those activities were conducted in partnership with the regional business community.

Mr. Beldona was the Larry Jones Fellow of Corporate Governance at Wichita State University from 2001?2007, where he was also the Entrepreneurship Research Associate for the school?s Center for Entrepreneurship. He was an associate professor of international business at the International University of Japan from 2000?2001, and was an assistant professor of international business at Rutgers University from 1994?2000.

He received a bachelor of science in physics and an MBA in general management from Karnatak University in Dharwad, India, and a master of science in international business as well as a PhD in business administration from Temple University in Philadelphia.

The Old Reader


As the name implies, The Old Reader (free) was designed in to imitate Google Reader before one of its major redesigns?specifically, the one that nixed social sharing in 2011. If you have long missed those interactive sharing features of Google Reader, The Old Reader is hands-down the RSS reader for you.

While stable, reliable, and fairly well designed, The Old Reader doesn't have quite as many useful features as the adept G2Reader, a PCMag Editors' Choice, although it does offer support for more languages (G2Reader has seven, and The Older Reader has 17).

The Old Reader works well, and, if you're migrating to it from Google Reader, it doesn't ask you to rethink how you use your feeds or what kinds of feeds you might want to import. Some other alternatives, such as Pulse and Taptu, take a magazine-like approach and encourage you to read content from big-name media outlets. They also push a flashy interface that's rich with graphics, which may not appeal to dyed-in-the-wool RSS junkies. In other words, The Old Reader is totally non-disruptive.

You can import your Google Reader OPML file to The Old Reader and see your feeds in the new service pretty quickly. While it did import a listing for my Google Alerts, I'm not sure how the app processed them, and they don't seem to be updating any more (the came through just fine in G2Reader and continue to update). In The Old Reader, my Google Alerts now seem to be affiliated with a service called PubSubHubbub. I'm not clear what that service is or what it does, but while I work it out, I can continue to receive Google Alerts through Google by turning them on for email notification.

The name of the game in The Old Reader is "social." Like Google Reader in the days of yore, The Old Reader lets you follow other users of the site and see what they share with the community. Likewise, users can follow you. You can connect to Facebook to publish feed items you like there, or save them to Pocket to read later, as both services are integrated.

One minor feature that I quite like sits off to the right side of the page, a "dead" notice that lists feeds that haven't been updated in at least three months. It helps me keep my feed list clean and up-to-date, although I'd love the ability to customize how long a URL or feed must be inactive in order for it to be declared dead.

In testing, I hit one snag that I'd call an out-and-out bug. When looking at a list of unread feed entries, if I jump down and open one that I haven't read yet, all the others above it get marked as if I had read them. That needs to be fixed.

The Old Reader doesn't have any mobile apps, and the design, while simple overall, looks a little busy due to how it displays some text. For example, green bubbles with white text at first glance appear to be tags, but turn out to be non-interactive labels. It's a minor point, although I wouldn't think twice to suggest The Old Reader is due for a visual refresh.

I love The Old Reader, but I must admit that I love G2Reader a little more. It includes a tool that automatically highlights in your feeds any keywords that you save in your settings, and looks sharper to boot. I was also impressed that it handled the importing of my Google Alerts, while The Old Reader left me befuddled on that front.

The Old Reader is hands-down the RSS feed reader to use if you crave social interaction. If not, go to G2Reader, our Editors' Choice, and give it a whirl. If you're more of a DIY person, definitely try CommaFeed, a wonderful solution that helps you build an RSS feed reader that you can host yourself. We also looked at 312914">Tiny Tiny RSS, an open-source platform also for building your own RSS feed reader, but found it painfully slow.

Facebook admits data breach exposed 6 million users

SAN FRANCISCO (Reuters) - Facebook Inc has inadvertently exposed 6 million users' phone numbers and email addresses to unauthorized viewers over the past year, the world's largest social networking company disclosed late Friday.

Facebook blamed the data leaks, which began in 2012, on a technical glitch in its massive archive of contact information collected from its 1.1 billion users worldwide. As a result of the glitch, Facebook users who downloaded contact data for their list of friends obtained additional information that they were not supposed to have.

Facebook's security team was alerted to the bug last week and fixed it within 24 hours. But Facebook did not publicly acknowledge the bug until Friday afternoon, when it published an "important message" on its blog explaining the issue.

A Facebook spokesman said the delay was due to company procedure stipulating that regulators and affected users be notified before making a public announcement.

"We currently have no evidence that this bug has been exploited maliciously and we have not received complaints from users or seen anomalous behavior on the tool or site to suggest wrongdoing," Facebook said on its blog.

While the privacy breach was limited, "it's still something we're upset and embarrassed by, and we'll work doubly hard to make sure nothing like this happens again," it added.

The breach follows recent disclosures that several consumer Internet companies turned over troves of user data to a large-scale electronic surveillance program run by U.S. intelligence.

The companies include Facebook, Google Inc, Microsoft Corp, Apple Inc and Yahoo Inc.

The companies, led by Facebook, successfully negotiated with the U.S. government last week to reveal the approximate number of user information requests that each company had received, including secret national security orders.

Female jury chosen for Zimmerman's trial

SANFORD, Fla. (AP) ? A jury of six women, five of them white and the other a minority, was picked Thursday to decide the second-degree murder trial of George Zimmerman, a neighborhood watch volunteer who says he fatally shot Trayvon Martin, an unarmed black teenager, in self-defense.

Prosecutors have said Zimmerman, 29, racially profiled the 17-year-old Martin as he walked back from a convenience store on a rainy night Feb. 26, 2012, wearing a dark hooded shirt.

The race and ethnicity of the minority chosen for the jury was not immediately available. Zimmerman identifies himself as Hispanic.

Two of the jurors recently moved to the area ? one from Iowa and one from Chicago ? and two are involved with rescuing animals as their hobbies.

One juror had a prior arrest, but she said it was disposed of and she thought she was treated fairly. Two jurors have guns in their homes. All of their names have been kept confidential and the panel will be sequestered for the trial.

The central Florida community of Sanford is in Seminole County, which is 78.5 percent white and 16.5 percent black, roughly mirroring the jury's racial makeup.

Prosecutors and defense attorneys chose the panel of six jurors after almost two weeks of jury selection. In Florida, 12 jurors are required only for criminal trials involving capital cases, when the death penalty is being considered.

If convicted, Zimmerman could face a potential life sentence.

On Feb. 26, 2012, Zimmerman spotted Martin, whom he did not recognize, walking in the gated townhome community where Zimmerman lived and the fiancee of Martin's father also resided. There had been a rash of recent break-ins at the Retreat, and Zimmerman was wary of strangers walking through the complex.

The two eventually got into a struggle and Zimmerman shot Martin in the chest with his 9mm handgun. He was charged 44 days after the shooting, only after a special prosecutor was appointed to review the case.

Martin's shooting death and the initial decision not to charge Zimmerman led to public outrage and demonstrations around the nation, with some accusing Sanford police of failing to thoroughly investigate the shooting.

The six jurors were culled from a pool of 40 candidates who made it into a second round of jury questioning. Two men and two women also were picked as alternate jurors.

Before selecting the jurors Thursday, defense attorney Mark O'Mara explored potential jurors' views on guns, self-defense and justifiable use of force.

Under Florida law, Zimmerman could shoot Martin in self-defense if it was necessary to prevent death or great bodily harm. O'Mara previously decided not to invoke a "stand your ground" hearing in which a judge alone would decide whether to dismiss the case or allow it to proceed to trial.

Lavrov: Syria peace conference could be derailed

ST. PETERSBURG, Russia (AP) ? Russia's foreign minister said Friday that Washington is sending contradictory signals on Syria that could derail an international conference intended to end the civil war, warning that U.S. talk about a possible no-fly zone would only encourage the rebels to keep fighting.

Sergey Lavrov, speaking in an interview with The Associated Press and the Bloomberg news agencies on Friday, also criticized demands that Syrian President Bashar Assad step down.

"Not because we like the regime, not because we want the regime to stay, but because it's for the Syrians to decide," Lavrov said. "And to say you must capitulate and deliver the power to us is just not realistic."

In addition, he dismissed allegations by the United States, Britain and France that Assad's regime has used chemical weapons.

Russia has been the key ally of Assad's regime throughout a two-year civil war, which has killed more than 93,000 people, shielding it from U.N. Security Council sanctions and continuing to provide it with weapons.

Russian President Vladimir Putin has confirmed that his country signed a contract for the delivery of S-300 state-of-the art air defense missile systems to Syria, but said it hasn't been fulfilled yet.

Lavrov defended the S-300 deal, pointing to the deployment of U.S. Patriot air defense missiles and fighter jets to neighboring Jordan.

"The contract on S-300s is absolutely legal, it's transparent and it's fully in line with the international norms and with the Russian export control legislation," he said.

"Second, the contract hasn't been yet finalized. Third, the Americans are leaving Patriots after this exercise in Jordan, together with F-16 planes, and no one is asking them not to do this. The region is really full of weapons, including offensive weapons which have been supplied in the past to the countries of the region, and some of these weapons are infiltrating into Syria."

He said supplies of weapons to the Syrian opposition, which have been promised by Washington and are being considered by the European Union, would be a "very big mistake."

Lavrov was asked if Russia is warning the West in particular against providing the rebels with shoulder-fired air defense missiles that could challenge Assad's air dominance. He replied: "We believe this is absolutely illegitimate, and we aren't going to legitimize it by starting discussing some conditions on which these supplies could be justified."

He said that while the U.S. says it favors sponsoring a Syria peace conference in Geneva, it has made statements that have sent a conflicting signal to the rebels. Lavrov said that the U.S. talk about a possible no-fly zone in particular has encouraged the opposition to step up fighting instead of sitting down for talks.

"The message the opposition is getting: Guys, don't go to Geneva, don't say you are going to negotiate with the regime, soon things will change in your favor," Lavrov said. "It's either the conference or the instigation of the opposition not to be flexible. I don't think it's possible to do both at the same time."

The date and location of the international conference on Syria haven't been announced yet, but it's already being dubbed "Geneva 2" since a similar event was held there a year ago.

"If our goal is the conference, then we must avoid any discussions and, of course, any action designed to establish a no-fly zone. We must avoid confrontational debates and one-sided resolutions in the General Assembly and in the Human Rights Council because all this isn't helping to create the atmosphere necessary to convene a conference," Lavrov said.

He shrugged off the U.S., British and French statements about the use of chemical weapons by the Syrian regime as ungrounded, saying they "smack of politics and speculation."

"We have been told by the Americans, by the French, by the British that they have proofs," he said. "What they showed to us is absolutely unconvincing. It's not based on facts, and it can't be taken as a proof."

He said that a new international probe must determine the truth and added that after the conflict is over, Syria could be encouraged to destroy its chemical weapons stockpiles.
